Ebenezer Scrooge (Jim Carrey) memang adalah seorang pria yang pandai berhitung. Tak ada hal yang ia lakukan sebelum melalui pertimbangan untung dan rugi. Suatu ketika Ebenezer Scrooge yang super pelit ini didatangi sosok gaib bernama Jacob Marley (Gary Oldman). Jacob memberi tahu bahwa Ebenezer akan didatangi tiga sosok gaib yang akan mengungkap kisah perjalanan hidup Ebenezer. Tiga sosok gaib ini akan mengantar Ebenezer melewati perjalanan panjang melampaui batas ruang dan waktu untuk membuka mata hati Ebenezer.

Yang muncul pertama adalah Ghost of Christmas Past (Jim Carrey). Ghost of Christmas Past ini membawa Ebenezer ke masa lalu untuk mengungkap apa sebenarnya yang membuat Ebenezer sangat membenci hari Natal. Setelah itu muncul Ghost of Christmas Present (Jim Carrey) yang mengantar Ebenezer melihat kebahagiaan dan penderitaan orang-orang yang ada di sekitar Ebenezer. Meski sudah meraih popularitas bukan berarti pendidikan lantas tak ada gunanya. Itulah yang dipikirkan David Seville (Jason Lee) yang mengirim Alvin (Justin Long) dan kedua temannya ke sebuah sekolah untuk kembali belajar, sesuatu yang tak pernah terlintas di benak Alvin.

Alvin and the Chipmunks adalah trio tupai yang pintar dan berbakat, terdiri dari Alvin yang nakal namun pintar, Simon yang sifatnya lebih dewasa, dan Theodore tupai gendut yang manja dan plin-plan.

Alvin, Simon, dan Theodore yang kehilangan rumah mereka, sebuah pohon cemara, yang ditebang untuk hiasan Natal. Sejak saat itu, kehidupan mereka pun berubah. Pohon yang terpajang di sebuah lobi gedung membawa ketiganya masuk dalam hiruk pikuk kehidupan manusia di Los Angeles. Akhirnya mereka 'terdampar' bersama Dave Seville (Jason Lee).

Awalnya Dave yang hidup sendiri, tidak menyukai kehadiran ketiga tupai usil pandai bicara yang mengacaukan dapurnya. Ditambah pula, Dave sedang kesal karena lagu-lagunya ditolak mentah-mentah oleh temannya, Ian Hawk (David Cross), pemilik Jett Records.

Namun di tengah keputusasaan, Dave menemukan harapan baru ketika mengetahui tiga tupai itu ternyata pandai menyanyi. Terinspirasi oleh mereka, Dave membuat sebuah lagu bagi ketiga tupai tersebut. Ia membuat kesepakatan apabila tiga chipmunk tersebut mau menyanyikan lagunya, maka mereka diperbolehkan tinggal di rumahnya dengan catatan mengikuti peraturannya.

Dengan sebuah perjuangan, akhirnya lagu itu diterima Jett Records dengan para penyanyi baru, Alvin dan kedua temannya. Dengan kesabarannya, Dave akhirnya berhasil mengantar trio chipmunk meraih sukses sebagai penyanyi.

Hal yang tidak mudah dilalui Dave, karena tupai-tupai itu sangat bandel dengan tingkat keisengan tinggi. Tingkah konyol mereka mengganggu semua segi kehidupan Dave, termasuk kisah cintanya dengan wartawati Claire (Cameron Richardson).

Hubungan Dave dan ketiganya pun merenggang seiring kesuksesan trio chipmunk. Ian yang tamak berusaha mengeruk keuntungan pribadi situasi tersebut. Ian pun membujuk dan membohongi Alvin dan kedua temannya sehingga meninggalkan Dave. Setelah itu Ian memanfaatkan ketiganya untuk kemakmuran dirinya. Kate adalah seekor serigala yang selalu memegang teguh tanggung jawab dan disiplin dalam segala hal. Kate tahu kalau hidup di alam bebas penuh dengan tantangan dan Kate tak mau berbuat ceroboh yang nantinya bakal mencelakakan dirinya sendiri atau serigala lain dalam kelompoknya. Alpha and Omega. The beginning and end. Ancient phrase is now represented by the figure of Kate (Hayden Panettiere) and Humphrey (Justin Long) who is almost diametrically opposed in every way.
